تولید و کاربرد Trichoderma harzianum Tr6 در کنترل مرگ گیاهچه ناشی از Phytophthora drechsleri و افزایش رشد در خیار

چکیده

جنس تریکودرما یکی از عوامل مهم بیوکنترلی در برخی بیماری‌های گیاهی است که با مکانیسم‌های مختلفی بیماری را کنترل می‌کند. این قارچ‌ همانند دیگر عوامل کنترل بیولوژیک تحت تأثیر فاکتورهای محیطی قرار می‌گیرد که محیط کشت از جملة این فاکتورها است. این تحقیق با هدف ارزیابی تأثیر ترکیب ملاس چغندر قند، شربت ذرت، مخمر نان و عصارة مالت (Mol.C.B.M.) به‌عنوان محیط کشت مایع، دانة گندم (W.s.) و سبوس گندم  (W.b.)به‌عنوان محیط‌های کشت جامد بر توان اسپوردهی، کنترل مرگ گیاهچه ناشی از Phytophthora drechsleri و توسعة رشد گیاهچة خیار توسط Trichoderma harzianum Tr6 انجام شد. محیط‌های کشت با 1 میلی‌لیتر سوسپانسیون اسپور آنتاگونیست با غلظت 108 اسپور تلقیح و سپس، در دمای 28 درجة سلسیوس و شدت نور فلورسنت 206 لوکس به‌مدت 10روز نگه‌داری شدند. تیمار مربوط به محیط کشت Mol.C.B.M. بیشترین توان اسپورزایی با میزان 1010×83/1 اسپور در هر گرم وزن خشک زیست‌تودة آنتاگونیست و نیز بیشترین درصد کنترل بیماری (5/62 درصد)، افزایش ارتفاع، وزن تر و خشک اندم‌های هوایی و ریشة گیاهچه را نشان داد؛ اما درصد بازدارندگی در شرایط آزمایشگاهی نسبت به محیط‌های کشت جامد کمتر بود. بنابراین، می‌توان گفت که محیط کشت مایع برای دستیابی به اهداف تحقیق، موفق عمل کرده است.
