مقایسة اثر محیط‌های کشت آلی روی میزان جمعیت تیپ وحشی و تیپ جهش‌یافتة باکتری بیوکنترل Pseudomonas fluorescens VUPf5

چکیده

در این تحقیق رشد دو سویة باکتریPseudomonas fluorescens VUPf5 و fluorescens VUPf5-1 P. (باکتری جهش‌یافتة مشابه جهش‌یافتة سیستم تنظیمی gac) در محیط‌های کشت آلی ارزیابی شد. این جهش خودبه‌خودی تولید ترکیبات ضد قارچی سیانید هیدروژن، آنتی‌بیوتیک فنازین و آنزیم پروتئاز را در سویة بیوکنترل Pseudomonas fluorescens VUPf5 متوقف کرد. با استفاده از محیط‌های کشت آلی همراه یا بدون سولفات روی سعی شد که جمعیت جهش‌یافته‌ها تا حد امکان کاهش یابد. با هدف افزایش رشد جمعیت VUPf5 و کاهش رشد جمعیت VUPf5-1، شش منبع کربنی (ملاس چغندرقند، نشاسته، سیب‌زمینی، خرما، شکر، سیب) ،سه منبع ازتی (سویا، اوره، سولفات آمونیوم) و یک منبع کربنی ـ ازتی (جوانة گندم) بررسی شدند. محیط کشت 13B (ملاس چغندرقند، جوانة گندم، سویا، شکر، سیب و سولفات منگنز)، محیط کشت بهتری از لحاظ جمعیت باکتری VUPf5 (1010× 56/5 سلول باکتری در هر میلی‌لیتر) در مقایسه با محیط‌های کشت دیگر شناخته شد و اختلاف جمعیت باکتری در مقایسه با سایر محیط‌ها معنی‌دار بود. محیط کشت 1D (ملاس چغندرقند، جوانة گندم، سویا، شکر، سیب و سولفات روی) سبب کاهش چشمگیری در تکثیر پرگنه‌های جهش‌یافته شد. نتایج به‌دست‌آمده می‌تواند مبنای قابل اعتمادی برای تکثیر بهینة عامل بیوکنترل در فرایند‌های فرمانتاسیون را فراهم کند.
