These bacterial isolates were screened based on their antagonistic potential against Rhizoctonia solani and their ability to form robust biofilm. Out of these isolates, 49 isolates exhibited more that 30% antagonistic activity against R. solani. Different methods were used to screen the isolates with high ability of biofilm formation. Out of 180 isolates, 51 isolates were able to form considerable amount of biofilm as determined by crystal violet staining (CVS) method. Pellicle formation (PF) bioassay was performed in three different media. Most of the robust biofilm forming isolates from CVS method could also form robust biofilms in PF method, only if MSNGP (medium with glycerol and pectin) medium was used. In root colonization bioassay, isolates N168, N95 and N94 that were robust or mediocre biofilm forming isolates in MSNGP, could efficiently colonize plant roots but the population of isolates N100 and N87 (none-biofilm forming isolates) on tomato roots were lower than 6 log10 CFU g-1 root. However, results of the pathogenicity system with R. solani damping off of tomato showed that, biofilm formation and colonization ability of these isolates doesn't have significant effect on their biocontrol activity}, keywords = {rhizobacteria,Rhizoctonia,biofilm,Bacillus,Pseudomonas}, title_fa = {The Impact of Biofilm Forming Rhizobacteria on Rhizoctonia solani Damping-off of Tomato Running title: The Impact of biofilm forming rhizobacteria on Rhizoctonia solani}, abstract_fa = {در این مطالعه، ۱۸۰ ریزوباکتر از مزارع مختلف استانهای آذربایجان شرقی و غربی جداسازی شدند. این جدایه‌های باکتریایی بر اساس پتانسیل آنتاگونیستی خود علیه Rhizoctonia solani و توانایی تشکیل بیوفیلم قوی غربال شدند. از بین این جدایه‌ها، ۴۹ جدایه بیش از ۳۰ درصد فعالیت آنتاگونیستی علیه R. solani از خود نشان دادند. روش‌های متفاوتی برای غربال جدایه‌هایی با توانایی تشکیل بیوفیلم بالا مورد استفاده قرار گرفتند. بر اساس روش رنگ آمیزی کریستال ویوله (CVS) از بین ۱۸۰ جدایه ۵۱ جدایه قادر به تشکیل بیوفیلم قابل توجهی بودند. آزمون تشکیل پلیکل (Pellicle formation bioassay-PF) در چند محیط مختلف به انجام رسید. بسیاری از جدایه‌های با قدرت تشکیل بیوفیلم بالا از روش CVS، تنها زمانی در روش PF بیوفیلم قوی تشکیل می‌دادند که از محیط MSNGP (محیط دارای گلیسرول و پکتین) استفاده می‌شد. در آزمون کلنیزاسیون ریشه، جدایه‌های N168، N95 و N94 که در محیط MSNGP بیوفیلم متوسط یا قوی تشکیل داده بودند، توانستند ریشه‌های گیاه گوجه فرنگی را به صورت کارآمدی کلنیزه کنند اما جمعیت جدایه‌های N100، و N87 (ایزوله‌هایی که بیوفیلم تشکیل ندادند) بر روی ریشه‌ها کمتر از 6 log10 واحد تشکیل دهنده کلنی به ازای هر گرم ریشه بود. هرچند نتایج سیستم بیماریزایی با مرگ گیاهچه R. solani گوجه فرنگی نشان داد که توانایی کلنیزاسیون و تشکیل بیوفیلم جدایه‌ها اثر معنی داری در فعالیت بیوکنترل آنها ندارد.}, keywords_fa = {ریزوباکتری,ریزوکتونیا,باسیلوس,سودوموناس,بیوفیلم}, url = {https://jbiocontrol.ut.ac.ir/article_72391.html}, eprint = {https://jbiocontrol.ut.ac.ir/article_72391_bddbf2dfca0e3a1ddab8e6f4625c34f7.pdf} }
