Recently, some soil inhabiting bacteria were isolated that degraded bacterial signaling molecules such as Acyl homoserine lacton (AHL). The aiiA gene is responcible for AHL lactonase enzymes in Bacillus species. In this research the effects of aiiA genes from Bacillus sp. DMS133 and Bacillus sp. A24 were evaluated on the motility properties and enzyme production in P. carotovorum. The results revealed that swimming, swarming and twitching motilities were significantly reduced in aiiA mutant pectobacteria (EMPCC/aiiA1 and EMPCC/aiiA2 strains) in comparison with the wild type EMPCC strain. The expression of aiiA genes in mutant Pectobacteria significanlty inhibited production of pectinase, protease and cellulase enzymes. According as the means of enzymes production in EMPCC twenty time more than mutant EMPCC/aiiA1 and EMPCC/aiiA2 strains. Motility and plant cellwall degradation enzymes are two most virulence determinants in Pectobacterium, which are regulated by AHL mediated quorum sensing. Heterologous production of AiiA lactonase enzyme in Pectobacterium causes degradation of signaling molecules and decreases accumulation of them around the bacterium. As a result, the quorum sensing and related phenotypes are inhibited in P. carotovorum.}, keywords = {Bacillus sp,heterologus expression,soft rot,virulence determinant}, title_fa = {تأثیر بیان ژن aiiA بر تحرک و آنزیم‌های بیماری‌زایی باکتری Pectobacterium carotovorum عامل پوسیدگی نرم باکتریایی}, abstract_fa = {حدنصاب احساس (Quorum sensing) یک مکانیسم‌ تنظیمکننده است که توسط‌ Pectobacterium carotovorum برای تنظیم بیان ژن‌ها و تولید فاکتورهای بیماری‌زایی استفاده می‌شود. در سال‌های اخیر باکتری‌های مختلفی از خاک جداسازی شده‌اند که می‌توانند مولکول‌های پیام‌رسان باکتریها به‌ویژه اسیل هموسرین لاکتون (AHL) را تجزیه و در حدنصاب احساس باکتری‌ها اختلال ایجاد کنند. در این پژوهش اثر ژن مولد آنزیم لاکتوناز‌ (aiiA) از باکتری‌های Bacillus sp. DMS133 و Bacillus sp. A24 روی تحرک و تولید آنزیمهای بیماری‌زایی باکتری P. carotovorum EMPCC مورد بررسی قرار گرفت. نتایج نشان داد که فعالیتهای حرکت سطحی(Swimming)، شناگری (Swarming) و خزیدن در عمق (Twitching) در پکتوباکتریومهای موتانت و دارای ژن aiiA (جدایه‌هایEMPCC/aiiA1 و EMPCC/aiiA2) نسبت به باکتری تیپ وحشی (جدایه EMPCC) به شدت کاهش پیدا کرد. در آزمون اندازهگیری آنزیمهای بیماری‌زایی نیز حضور AHL لاکتوناز در باکتری موتانت باعث کاهش چشم‌گیر مقدار آنزیمهای پکتیناز، سلولاز و پروتئاز شد. به‌طوری‌که میانگین تولید این آنزیمها نسبت به باکتری فاقد ژن aiiA تا بیش از بیست برابر کاهش نشان داد. تحرک و تولید آنزیمهای تجزیهکننده دیواره سلولی گیاه از فاکتورهای مهم بیماری‌زایی پکتوباکتریومها هستند. حضور ژن aiiA لاکتوناز در این باکتری باعث تجزیه شدن مولکولهای پیامرسان و در نتیجه اختلال در فعالیتهای وابسته به QS به‌ویژه فاکتورهای بیماری‌زایی در P. carotovorum شده است.}, keywords_fa = {Bacillus sp,بیان ژن aiiA,فاکتورهای بیماریزایی,پوسیدگی نرم}, url = {https://jbiocontrol.ut.ac.ir/article_70620.html}, eprint = {https://jbiocontrol.ut.ac.ir/article_70620_a6ce8c43c6876637839c9f7a76af41ee.pdf} }
