This study aimed to isolate, identify and evaluate the antimicrobial activity of Streptomyces species in Oman Sea against three plant pathogens including Fusarium oxysporum, Xanthomonas campestris and Erwinia amylovora. The Streptomyces strains were isolated on different media including starch casein agar, glycerol glycine agar and starch nitrate agar with different NaCl concentration. Preliminary antimicrobial activity was investigated using cross streak method and evaluated by measuring the zone of growth inhibition against plant pathogens. Ninety percent of the strains were able to inhibit the growth of X. campestris strain and the highest inhibition zone was 15±1 mm. Two strains, AC275 and AC130 were selected to extract the antimicrobial compound. The isolates were cultured in different seed cultures and fermentation media. The application of different fermentation media shows that glycerol and soybeans cultures increase inhibition zone. Based on 16S rRNA gene analysis, two strains were belonged to Streptomyces sp. The organic solvents including dichloromethane, chloroform and ethyl acetate were used in order to extract the antimicrobial compound. Dichloromethane in AC130 and chloroform in AC275 were more effective than other solvents by creating an inhibition zone of 21 and 25 mm respectively. This study shows that the Streptomyces species in less explored marine environment have the ability to control plant pathogenic bacteria.}, keywords = {Antimicrobial activity,Marin Streptomyces,plant pathogens,sea Oman}, title_fa = {بررسی خصوصیات ضد‌میکروبی برخی سویه‌های Streptomyces دریایی بر علیه سه بیمارگر گیاهی}, abstract_fa = {در سال­های اخیر، استفاده از متابولیت‎های ثانویه مانند آنتی‌بیوتیک‌ها به ­منظور کنترل زیستی عوامل بیماریزای­ گیاهی مورد توجه قرار گرفته است. این پژوهش با هدف جداسازی، شناسایی و بررسی فعالیت ضدمیکروبی جدایه‌هایی از Streptomyces از دریای عمان علیه سه بیمارگر گیاهی Fusarium oxysporum، Xanthomonas campestris و Erwinia amylovora اجرا شد. سویه‌های Streptomyces روی محیط کشت‌های نشاسته کازیین اگار، گلیسرول گلیسین آگار و نشاسته نیترات آگار در غلظت‌های نمکی متفاوت جداسازی شدند. فعالیت ضدمیکروبی سویه‌ها با استفاده از کشت تقاطعی و اندازه‌گیری هاله عدم رشد در برابر بیمارگرهای گیاهی اندازه­گیری شد. 90% این سویه‌ها قادر به تولید هاله مهارکنندگی رشد در برابر باکتری X. campestris بودند که بالاترین هاله 1±15 میلی‌متر به‌دست آمد. دو سویه 130AC و 275AC برای استخراج ترکیب ضدمیکروبی انتخاب شدند. این سویه‌ها در محیط­های پیش­کشت و تخمیری مختلفی کشت داده شدند. استفاده از محیط‌های تخمیری مختلف نشان می‎دهد که محیط دارای گلیسرول و سویا هاله بازدارندگی را افزایش می­دهد. سویه‌های به‌دست آمده براساس ژن SrRNA16 متعلق به گونه‌هایStreptomyces sp. می‎باشند. جهت استخراج ترکیب ضدمیکروبی از حلال آلی دی‎کلرومتان، کلروفرم و اتیل‌استات استفاده گردید. حلال دی‌کلرومتان در سویه 130 ACو کلروفرم در سویه 275AC با ایجاد هاله مهارکنندگی 21 و 25 میلی‌متر مؤثرتر از سایر حلال‌ها بودند. این مطالعه نشان می‎دهد که برخی جدایه‌های Streptomyces در محیط‎های دریایی کمتر کاوش شده، دارای قابلیت کنترل باکتری‎های بیماریزای گیاهی می‌باشند که می‌توانند مورد استفاده قرار گیرند.}, keywords_fa = {Streptomyces دریایی,بیمارگرهای گیاهی,دریای عمان,فعالیت ضدمیکروبی}, url = {https://jbiocontrol.ut.ac.ir/article_62758.html}, eprint = {https://jbiocontrol.ut.ac.ir/article_62758_104f3dbbf40ff530d4fb6f134929dd0a.pdf} }
